Formulation and characterization of mucoadhesive bilayered tablets of diclofenac sodium using various polymers

The purpose of this research work was to establish mucoadhesive buccal tablets of Diclofenac Sodium in the form of bilayered tablets by direct compressionmethod. The tablets were prepared using Sodium alginate (SA) Sodium carboxymethylcellulose (SCMC) and Hydroxy propyl methyl cellulose (HPMC) asbioadhesive polymers to impart mucoadhesion and ethyl cellulose (EC) to act as an impermeable backing layer. Buccal tablets were evaluated for differentparameters such as weight uniformity, content uniformity, thickness, hardness, surface pH, swelling index, ex vivo mucoadhesive strength, ex vivo mucoadhesiontime, in vitro drug release, and in vitro drug permeation. The mechanism of drug release was found to follow Zero Order drug release with non-Fickian typediffusion (value of n between 0.5 and 1.0) for the best formulation. The stability of drug was tested for 6 hours in natural human saliva; all the buccal tabletswere found to be stable in natural human saliva.The present study concludes that mucoadhesive buccal tablets of Diclofenac Sodium can be a good way to bypassthe extensive hepatic first-pass metabolism and to improve the bioavailability and duration of action.
